First of all, she's well aware of her many flaws: a tendency to panic, poor (though gradually increasing) social skills and ineptitude at first meetings, nervousness and a tendency to panic and overreact, and most cripplingly, apparent obsessive-compulsive personality disorder. But she's also capable of amazing strengths: she's incredibly talented at magic (as the cutie mark on her flank shows), and more importantly she has come to understand friendship and can use her faith in her friends to not only increase her power but to weather even the most terrifying of situations. Faithfully studious, she's a major bookworm and is frequently considered endearingly awkward in her enthusiasm about the real world and friendship.
Life Story (as much as you want us to know): Once upon a time in the magical land of Equestria... Well, let's cut to the chase. Twilight Sparkle was a privileged but shy unicorn filly growing up in the wealthy capital city of Canterlot, where she had a brother but no real friends at all. She came to idolize the all-powerful god-princess Celestia, and threw herself into her studies in an attempt to master the arcane arts. After an...incident, she found herself studying for years as Princess Celestia's personal student, and with a new assistant/little brother figure, Spike the baby dragon.
Eventually, just as she became convinced that Celestia's sister--turned evil and banished to the moon one thousand years ago--would be coming back, Celestia seemingly inexplicably sent her to Ponyville, a provincial town with a strange assignment to study friendship. It turned out for the best: after making five best friends, she found that they were the chosen ones who would once again wield the Elements of Harmony, powerful artifacts that allowed them to return Princess Luna to goodness.
After saving the world from monstrous beings of ultimate power several more times, and more traumatic yet, attending her brother's wedding, Twilight was granted ascension to alicorn (winged unicorn and ultimately powerful being) status. And then, one more incident involving a human world occurred, and the next thing she knew she appeared in an entirely different city...as a unicorn again. Stranger in a strange land, as ever.
